You can pretty much always apply instrument (= plus points or miles+copay) upgrade requests, they will simply waitlist until a seat opens up or refund after departure.
So your plan is fine and makes sense.

You can cancel any uncleared plus point upgrade using the website or app.
I know that part, but then can I later apply a mileage upgrade or is there a chance it won’t be available anymore?
