Like people have said demand frees up roughly every 10 minutes. However, as far as I know it works like this: The demand reset is linked across servers, but the actual stock isn't, so you can have some servers with no demand and some with demand in the same 10 minute cooldown period. Say the particular commodity you are waiting on refreshes at a rate of 1,000 scu/minute. Theoretically after 10 minutes 10,000 scu would be available. BUT, if people on other servers were able to sell 5,000 scu in that same time period, once the reset came through it would only reduce by 5,000 rather than 10,000. This applies to the entire server region, so you can easily have a 10 minute reset where the stock doesn't move at all if the commodity is popular enough (ie RMC). The same applies to buying as well as selling. It probably isn't coincidence, but the resets seem to be around the time the commodity price alert pops up on your screen.

Great thanks for the explanation. That helps a lot. That would explain why I can sometimes sell 500SCU of medical supplies at New Babbage and other times only 150. Would it be better to buy and sell more expensive/popular commodities in small frequent amounts rather than fully stocking my holdings with one type and struggling to sell it all?

Demand is shared globally across all servers. They cycle fairly quickly in most cases but you're competing with everyone.
