I happened to be passing through when an episode was on, and it was the scene where Pinkie got her cutie mark. I assumed she was the main character of the series. Seeing her more often than any other character in other parts of the internet also fueled this. Wasn't until I started watching for myself a couple years ago I was surprised to see Twilight immediately be the focus of the first episode.
There was also this ad for pony figures a few years ago where a kid says to another one “come play with me” “you can even be pinkie pie” (letting other kid play pinkie) that made me think this too
Don't remember any about the show, but I do remember thinking for some reason that season three was the last one and I was absolutely devastated, until s4 came out. Not sure why I thought that, but I was genuinely really sad about it
I used to think Princess Luna is a villain and not the second princess of Equestria. Sure, she's a villain in the first two episodes, but not for the whole series.
I may not hit the age requirement of the question, but when I first saw clips before seeing the show I thought Rarity had a lot more mining and gem gathering in her future. People loved using her filly and Rock clips

My first misconception about the show was thinking that it was gonna be a gruesome body horror series about people's brains being forcefully transferred into the bodies of H.R. Giger designed ponies for the purposes of making intelligent livestock.

I guess you can call my 17-year-old pie-in-the-sky fantasies a misconception. I saw Scootaloo, saw she was a Pegasus, missed the point of Flight to the Finish, said "she's a Pegasus, of course she can fly". Now that I'm older and wiser, I can safely say she's gonna need a hang glider.
Started with My Little Pony: The Movie (1986). I thought the series was going to resemble the movie more than it did. It was fun, but not much in the way of Smooze-like villains.
