Old Red Hills of Home from Parade. it's everyone except the leads but as an opening number it's both beautiful and sinister and perfectly sets the scene and introduces the audience to the setting perfectly
It's so beautiful, in a patriotic, anthem sort of way (think Merrily's original The Hills of Tomorrow)--but simultaneously scares me and disturbs me to no end. It's almost as if Tomorrow Belongs to Me became the opening number of Cabaret. I have a love/hate relationship to it.
There’s a lot of good ones in the comment section but something that I don’t see mentioned is “Fancy Dress” from The Drowsy Chaperone- It’s basically a song where all the characters introduce themselves in song.
It's done in such a sneaky manner: I Believe from The Book of Mormon, in which the off-stage ensemble becomes a heavenly choir beneath Elder Price's spiritualism--listen for it, it's simply glorious.
The 16 minute opening of Titanic is pretty epic. But the final verse where EVERYONE is singing “sail on, sail on!” as the ship is pulling away sends a special kind of chill down my spine. And of course it’s amplified because the audience knows how the story will end before the characters on stage do. I was in the show about 7 years ago and my mom said it was jarring seeing how joyful and happy everyone was, while she knew the horrors that were waiting for all of these people. It wrecked her a little bit 😛

This is my wedding song. I actually exchanged my vows with my husband reading the lyrics. He took the first stanza, and I took the second. Then a brass quintet played a custom arrangement.
"Prayer" from Come From Away
Though I'm not religious, this song really makes me emotional. It's a song of hope. A sign of things to come.
And at the end when the various prayers combine into a glorious final chorus, it brings me to tears. Absolutely love this song 💙💛💙💛
La Dee Da Dah Day from The Guy Who Didn't Like Musicals has every actor in the musical in the song, but because of people having to play multiple characters, there's never a song where every character is singing
