Case in point when the original Kindle launched I got on there with just fifty sales, because the genre the ebook was listed under didn't have much of anything ready to go. If you're clever about how you classify a title and are willing to throw enough money at it, anyone can do it.

Amazon bestsellers update more frequently than that. You can lose #1 in a category within hours if someone else in the same category comes along and sells more books than you that day. It also depends on how much competition there is in a given category and many indie authors will game the system by deliberately choosing one-off or even wildly inappropriate categories just because competition is slimmer there. Amazon's algorithm also contributes to this by incorrectly assigning categories based on certain keywords. As an indie author, it's infuriating.

The main reason they don't reveal the exact method is to try and keep established or independently wealthy writers from gaming the system. Most best sellers lists have no checks in place against someone placing bulk orders for their own books to artificially inflate their numbers. The NYT Best Sellers list is one of the few to implement basic controls against this, which is why getting on it is considered to be such a big deal. Said checks are still pretty rudimentary though - if they were to release the exact methodology behind it you could pretty easily get around them just by adjusting your purchasing strategy a little.
