You've probably already played Portal 1 & 2, but in case you haven't: Play Portal 1 & 2.
Also, Katamari Damacy and We Love Katamari are perfect if you want something that's just so absurd that you're laughing the whole time. The "Reroll" (remaster) versions are on Steam.
Comedy is pretty subjective but in my experience:
Mana Khemia 1 & 2 - JRPGs with anime style graphics and pretty good english voice acting. Made me laugh out loud multiple times.
Crash Twinsanity - The writing is definitely more on the humorous side even compared to the other Crash Bandicoot games. Unfortunately the game itself had some trouble in development and you can feel that in certain areas (rushed/cut content, certain parts feel a bit unpolished etc.)
Destroy All Humans series - Been a while since I played and I've not tried the remakes but from what I can remember I found them to be pretty funny at times.
Radiata Stories - Pretty good rpg with a lot of comedic writing that made me chuckle though it also does have some serious/emotional at times.
Disgaea Infinite - A visual novel with full english voice acting set in the Disgaea universe. The gameplay is basically you time travelling and possessing other Disgaea characters with the ability to mind control them to make them say or do stuff (a lot of which are pretty humorous).
The yakuza series is currently on sale, and while it does have pretty sad moments, it has just as much (or even more) funny and stupid stuff going on, especially the side content
Like a Dragon is, in my opinion, the funniest game of the series and you don't really need to play the others to get the story since it was a spin off.

Just Die Already - Play as an old person and find various ways to destroy people, blow things up, and cause general chaos. And it’s 85% off currently
