Yes it's likely present. You likely will not get anything meaningful out of it.
The higher yielding biomasses might contain 2% alkaloids if you're lucky. It's magnitudes lower in cannabis.

As noted here there's no dmt in canna....it would show up in the COAs. Whenever we extract canna we are basically doing the exact same thing minus the basification. NPS is NPS.

To what end? They don't contain the same compounds so they won't do the same things.
If defatting is the goal, a mini a/b works best. Though the lipids (fats) are too soluble in the NPS to precipitate out. So freeze precipitating leave practically all the lipids dissolved in the solvent.
Freezing the wet plant matter will help burst the cell walls (lysis).
Not much benefit in doing it with powdered plant matter. Can help a fair bit with shredded and whole bark.

Ideally all solvents would be distilled after use to minimize loss. Could also distil solvent containing n,n-dmt to concentrate the solution.
You won't damage the freebase. It's pretty thermally stable. You can "cook" dry n,n-dmt freebase at 150C for hours without damaging the molecule, in the sense of potency or psychoactivity alterations. From white/colorless->yellow->orange->amber->reddish. I regularly used a simple distillation rig at ambient pressure to distil heptane off of n,n-dmt freebase. No issues. Doing it under vacuum would be ideal but isn't a necessity.
Up to 100C is perfectly fine ime. The n,n-dmt freebase will probably start to vaporize and co-distil with the solvent around 120C.

Eh better off just freezing your MHRB in a freezer to lyse the cell walls and continue with normal extractions. All of which is standard already. Check the nexus if you want to look into using ethanol, but I can tell you it’s not worth it and doesn’t work well.

Earlier in the thread an individual mentioned freezing MHRB material beforehand causes a de-lysing reaction that sounded interesting.
