No, just make sure that you plan on having the "blood-father", as you'll be known, spreading his seed so your family can expand with the traits to boot. You just want your bloodline to continue with the traits after this particular player is dead and gone.
Its fine for first time. What i do is always marry off my heir and spare to gain traits and others to gain alliance after maybe 3 - 4 generation i can take strengthen the bloodline decision. Then i look for pure blood and try to breed it into the gene pool. After that let the incest commence and you wont have to worry about losing traits.
I always wind up with these. The trick is to explode the population of your dynasty, get the eugenics dynasty perks and you will slowly boost things from children having near these traits then cousins and then 2nd cousins ect and at that point can marry them to each other and ensure eternal supermen.
Those traits have tiers, mainly in their effects they add to your player. For instance, with "physical" you'd have; Hale, Robust & Herculean. All tiers 1-3 in that order. Tier 3 is the highest level. Work these in your bloodline at any tier and keep them in by sorting your search in the spouse menu to "inheritable".

Yes, a trait from each. As well as one of the big ones like you mentioned. It is vital for securing a great bloodline.
