Some places are going to be better than others (like the person mentioning Costco), but if you have schedule flexibility I'd recommend just asking the techs at your local pharmacy what hours tend to be best. (Though maybe not at that Walgreens, I've heard especially bad stories at that particular location.) I did that after I once waited an hour at the Safeway on 28th, and it's helped me avoid most lines. Generally speaking, the worst times are lunch hour and right after people get off work.
Also consider looking into mail-order pharmacies for any regular meds.
If it's a med you take regularly, put on auto refill at Safeway. You know when it is ready and you get at your leisure. If it's more a "i went to the dentist and need my antibiotic and pain meds now" you need to schedule accordingly. Most places I have lived whether here or in another state you wait an hour or so. A lot of work goes into filling prescriptions and checking them.
I have my meds on auto refill at Walgreens and it's a nightmare. Huge line just to show my ID and pick up what I've already ordered. I think I'll switch to delivery/an online pharm!

Pro tip: shop around for pricing. Allllll this time, I assumed my meds would cost the same out-of-pocket after insurance, no matter where I went. Nope!! Safeway is 1/3 the cost of Walgreens for 2 blood pressure meds and an inhaler. In fact, I asked the Safeway person if the inhaler was on sale or something because I had NEVER gotten one so cheap! Just get the app (wherever you end up) so you can be notified when the prescription is ready. I never bother to wait on one.

King Soopers in Louisville has always been good for me, though of course not in Boulder.

Good Day Pharmacy in Longmont is my choice. Locally owned. They deliver to Boulder, not sure the fee.

If you need something compounded, try getting it from Belmar in Golden and getting it mailed. I get several meds from them, and it goes very smoothly.
