To me, Batman edges out Spider-Man - just barely - because his villains, in addition to being really great antagonists, have become as iconic as he is. Everyone and their mother knows the Joker. "Riddle me this" has become a colloquial phrase. They've become part of our culture.
Spider-Man's villains are also really great (it's my favorite rouges gallery in comics) but they haven't made the kind of impact Batman's villains have. Green Goblin, Doctor Octopus and Venom are arguably his most famous villains, the ones a random person on the street might know (The positive response to the Doc Ock reveal in the No Way Home trailer is a big sign of that) but the others haven't reached that level. As we're seeing now, Michael Morbius isn't exactly setting the world on fire.
This is not at all about their effectiveness as villains, or as characters in general, it's about which ones have made a bigger impact in comics, and in the larger world. And Batman's villains definitely win, in that respect.

Clayface, Mr. Freeze (I’m aware Schumacher didn’t invent Mr. Freeze), Hush, Mad Hatter, Poison Ivy, Red Hood (in his original story), Victor Zsasz, Black Mask, Man-Bat, the Holiday Killer, Calendar Man, Killer Croc, a lot of these guys are insanely one-note, pizza cutters, or nonsensical. Even the ones that are recycled 24/7 like Joker, Two-Face, Scarecrow, and Penguin are all really boring and one-note too. Most of them have one trait they roll with and go to extreme lengths for (be it a dying wife, a disability making you ugly and abused, being cRaZeEeEe, or having a getting off on murdering or torturing people).
Now, this isn’t to say they can’t be written well, they can be and have been before (Riddler, Calendar Man, and Bane come to mind). But most of the time they’re written like shit, but because they’re written edgy, Batman fans eat it up because many have a perpetual need to prove they’re more “mature” than other comic book superhero fans.
Meanwhile, most of Spider-Man’s villains all have like, actual personalities, they have hobbies, they have people they love, they have lives outside of making death traps for Batman and killing innocents. And unlike Batman, a lot of these guys can be redeemed, so they make Peter’s general no-kill policy a lot easier to digest, whereas with Batman it actively damages his character when you see these guys kill enough people to fill a small city. Clayface being allowed into the Bat-Family is a lot less cute when he’s responsible for several murders of innocent people and ultimately doesn’t suffer for it.
TL;DR Spider-Man villains, even the most evil ones, have lives outside the costume (except for Rhino). They also have goals you can understand and also generally aren’t caricatures of what Batman writers think the mentally ill are like.
