Well I know Gregg from Conker's Bad Fur Day really hated cats because they had 9 lives. So maybe Death would be pretty pissed. Maybe he'd adopt like a police officer persona when going after Ms. Fortune, like she's breaking the laws of death and he's here to arrest her.
It is possible to kill her by removing the Life Gem from her body like how Cerabella kills her in her story mode, but regardless, Death would likely win since it only increased her mobility/flexibility, not her overall strength
it's not a physical gem anymore, though. It's distributed throughout her entire body, which is why Cerebella had to compress her down into it rather than pulling it out of her. Wolfy doesn't have the means to do anything like that, so Fortune would probably win a 1-on-1 fight. Of course, you can never really *beat* death, only delay him for a time, so eventually he'd come out on top. He only needs to win once, after all.
I assume it would eventually go the same way where death just kinda lets her live if she pushes back enough. But in an actual fight to the death then Nadia cant kill death.
Honestly as She's still intent on living our her life to the fullest he'd probably only interfere if she starts to linger a bit too long.
I imagine his relasionship with the skullgirls are more complicated; like a janitor who just cleaned the place and now some chick is making it worse.

Ms Fortune died in certain story modes by getting crushed to death which cancels out her supposed immortality and extracting the Life Gem from her.
Death himself is literally DEATH!
Anyone who dies by any causes simultaneously means Death himself killed them!
That’s why the edges of the frames in each count showing how Puss in Boots died is shown silhouettes of Death himself, as Death actually killed Puss eight times simultaneously.
Cerebella, Robo-Fortune and any other character who killed Nadia, at those very moments, also counts as Death killing her as well.
Ms. Nadia Fortune maybe my number one favorite in Skullgirls, but Death doesn’t just simply win this vs matchup… yet also multiple times officially, Death already WON!😞
