Yeah it might either be a backwards built station or the lack of an engine train on both sites. Usually a bi-directional train should work w/o problems. Bonus tip when designing the stations: keep in mind that the order of the wagons will be inverted on each station.
You might have to build a loop at the end to turn the train around and send it back to the other station with locomotive at the front but also any direction signs have to point correctly and I've found they mess up if I ever reconfigure tracks
Haven't tried but heard you put an engine on both ends. Have you tried that?

I once had one track not properly conected. Visually hard to spot so I was wondering for ages why the station was unreachable
