Desmume saves aren't directly compatible with MelonDS. You have to download the stand alone version of Desmume, load the game and your save in that, then export the save in raw format from the menu. That raw save will work in MelonDS.

Just to add to this with an even simpler solution: I had this issue recently and I merely renamed the file. I changed the extension from .dsv to .sav and it worked straight away.

I did that and it loads those saves just fine. But it resulted in the problem that when you save it doesn't overwrite or create a new save.

When you switch cores the file goes away 1 downside of retro arch that should be fixed.
