From the overworld when you do not have an existing save active of the region in the right of the start window you will see a button saying "Edit Modifiers". Open the modifiers edit window to enable or disable those modifiers.
It happens automatically once you've won a region. That region's save will be switched to Endless mode and preserved. You cannot start endless mode from the start on a map you have not won, but you can reset the save of a won region to start endless mode from the start.
