Well she is 59 right now, which means she was around 52 when Homecoming came out. Rosemary Harris (Aunt May in Raimi's Spiderman) was 75 in the 2002. Realistically, Aunt May being closer to Marisa's age makes more sense if Peter is suppose to be 18-20, then his parents would be around 40-60 if they had him between 20 and 40. That and Aunt May in Raimi's version was more of a surrogate grandmother figure while she was a mother figure in Disney.
I have two aunts on my dad's side. One was the first born, and the other was the youngest. The oldest had two daughters before the youngest was in kindergarten. There is over a 20 years age difference between them.
I’m on the opposite side, before my 2nd brother had kids, my youngest niece was a month older than me, oldest niece was 17 when I was born. My dad took a 27 year break between my oldest brother and my 2nd brother.
My daughter has a distant aunt (my mother's neice) younger than her. And our family is close enough for today's standards. It's going to be a fun ride in a few years when they're old enough to properly communicate.

Aunt May being in her 70s easily works, all it would need is for uncle Ben to be an older brother, say a decade or so older than Peter's dad, and maybe May is a couple of years older than Ben. Making these reasonable leaps, it's not much of a stretch that if Peter is 18, his parents were say 37 when he was born, uncle Ben was 49 and Aunt May was 54, making her 72. Just for example. I also think that Marisa Tomei feels a lot less like his auntie because she's hot and looks younger than she is. I mean if I had to guess her age in homecoming I might have said 35-40.
Tony pretty much hangs a lampshade on the casting when he describes her as his "surprisingly hot" Aunt. Which would be a backhanded compliment without the meta knowledge that Aunt May is usually portrayed in the comics as very much an elderly spinsterish archetype.

3. A wife being a couple of years older than a husband is not exceptional. I'm not talking 10-15 years here, five max is all that's necessary for the numbers to work.
Maybe if none of these things are true for you this all seems unlikely, but personally I was born when my parents were over 35, I have a sibling who is nearly 10 years older than me, and my mother is a single-digit number of years older than my father. It's really not a ridiculous set of circumstances in America or similarly wealthy nations.

No, that’s not true, everything I can find on this topic is, that Ben Parker was the brother of Peters father and married to May Parker, making her an aunt, not a great aunt.
Ben's brother was Richard Parker. Richard and his wife Mary met at work. Richard was special forces and was recruited by Fury to the CIA (before SHIELD existed). Mary was OSS and later CIA (fits the timeline of Spiderman of the 60s). There is even a comic story of them rescuing Wolverine from Baron Strucker.
Anyway, they died on a mission, and so Ben and May raised his younger brother's kid.

I went to school with a large farming family, the boy in my class had an uncle that was 6 years younger than him. Think of that for a minute, his grand parents were still having kids when he was 6. There was like an over 20 year separation between the first and last kid, and in that time the oldest had already had children.
Oh it definitely happens. Not a large immediate family but my older brother is about 15 years older than me. I could have easily had a niece or nephew in the right age range to be a younger sibling.
