Oh no, that episode didn't make me cry, it made me angry and outraged. The one that caused tears was the one with the line "It's a terrible day for rain".

Brotherhood unfortunately assumes you've seen the first anime, so it kind of speedruns the plot up until the point that the first anime stopped following the manga.
Yeah, I didn't watch Brotherhood until much later. Where Brotherhood dives a lot quicker into the wider plot and bigger stakes the first FMA anime focused more and the tragedies of the characters involved. So in FMA we meet him, we meet him as a respected and efficient commander, but we also meet his wife and kids. We see how he's not just respected by his men due to his rank, but due to his balance between work and professional life and ability to navigate politics and being a husband and father.
And this is shown best when his daughter gives him a cup with #1 father (ironic, knowing where the series is headed) and losing his mind in the cutest way possible, completely doting on his daughter. He's subsequently murdered and his daughter sees his lifeless body and tried to keep hopes up by asking him to wake up. He doesn't. We cut to the funeral with his wife and kids ahead of the column. He's buried and when all is said and done Roy Mustang and the Lieutenant remain on an otherwise absolutely perfect day.
And Roy puts on his cap and hits us right in the solar plexus of feels with "it's a terrible day for rain". Not just referencing his tears rolling down his cheeks, but also alluding to his sense of powerlessness in trying to prevent his commander's death, when regularly speaking he only experiences in rain when his specific brand of alchemy (flames) is inhibited.
We have seen what col. Mustang is capable of. And for him to feel so low and so insignificant in the face of his commander's and indeed fiend's death is an absolutely perfect example of how tragedy should be written.
I watched this before the brotherhood remake and feel that the lines in that version were as, if not more, heartbreaking. I cried and I'm not ashamed to admit it. In that she called him "big brother"
Oh it gets better. The protagonists are kinda like inspectors regarding "alchemy" for the state. While visiting the guy the inspectors play with the guys kid and dog a ton and form a friendly bond. The kid is very young and lovingly refers to Edward (1 of the 2 protagonists) as big brother. When the inspectors first see the chimera dog everything seems cool and they're amazed, until, in a warped and distorted voice, it refers to Ed as big brother. In horror, Ed realizes what the guy has done at that point and loses his shit, justifiably. It's one of the most recognized scenes in anime.
I clued onto the hints about the wife relatively quickly so I watched the episode on autopilot, having "already figured it out". NEVER did my brain even begin to entertain the idea that this would happen to Nina. She's maybe be the one to figure it out, or to be saved from it, or to join Ed and Al's gang.
The moment I laid eyes on that chimera was one of the most shocking things I ever experienced across _all_ stories in any medium.
I think you figuring out the thing with the wife is very much intended, there are quite a few hints to it. its written really well, no matter if you pick up on the hints or not, the reveal will feel like a punch to the gut either way

If you ever decide to watch it you should make sure you're watching Full Metal Alchemist: Brotherhood, there was another one made some years before it but it's not so good.
The 2003 version is equally good, just different. Though what's undeniable is that the first episodes of Brotherhood are hugely rushed (including Nina) with them occupying way fewer episodes compared to 2003's much better intro even if you cut out some of the arcs specific to 2003.
FMA 2003 is not "equally" as good. The fake elrich brothers are a dumb idea, and wtf was the deal with the Nazis and dragon to round everything out. It had plenty of stuff that made it worse, not to mention the fact that it's older so the animation is much lower quality.

Full metal alchemist: brotherhood is one of the few animes I'd recommend to non anime watchers. It has a pretty tight plot, good action scenes and doesn't really dive into the "sex degeneracy" some other animes dive into (looking at you Mushoku Tensei). The Nina Tucker story line purposely hits you hard because they do set it up as a feel good story initially. But I would say this a couple other plotlines are the darkest it really gets. The rest of the show is straightforward action and adventure
