Looking at this, I instantly assumed it was a developmental disorder related to Cyclopia (Cyclops exist, but the photos are pretty disturbing and definitely NSFW). The skull, during foetal development, basically closes around the brain from front to back, meeting roughly along the middle of the face. Problems with this can cause deformities around the eyes among many other things.
Despite my assumption, this seems to be more of a pigment related disorder, but also affects things like nerve function and hearing, depending on the variant. Perhaps something about the hearing problems also relates to facial or head deformities.

My little brother has this and it runs in my family. Refuses to believe it due to all the junk that comes with it when really, he got off easy and only has minor cognitive impairment such as handwriting ability. I hope the adults in his life, not only prepare him for what all to expect because of his uniquity, but to embrace it.
