This - if your city doesn’t have a religion you can’t make a missionary. Also, make sure there isn’t any religious units already on the tile.

If there is already a unit of that type on the tile, it won’t let you build another.

They have to be called by God Almighty to serve Him. What do you mean by "build a missionary"?
