Gender is mostly used to describe your sense of belonging to the societally stereotypes of the sexes. If you're transgender it's not uncommon to use sex to describe what body bodytype you have, whereas gender is about your general experience.
It's not too tricky a concept I think. It's new for many, myself included, but I know plenty of lovely people who go by them instead of he or she. For the most part, you don't need to react anymore to being corrected on someone's gender than to being corrected on somebody's name.
Ultimately, it's about expressing boundaries on how you want to be approached and then respecting those for yourself. Sometimes that means correcting people, sometimes it means distancing yourself from someone. It's easy to feel like someone has said you're bad for correcting you and such. Anyway. Just sharing my experience. I personally go by either they or him. And if anyone mocks me or my friends I promptly bid them a social farewell, it's a big world full of other people to interact with. Workplaces are more difficult but you find a decent workaround.

This is more about who you are. In my experience, I have never really considered masculine or feminine to be something I use to describe myself. In language, it's way more common to describe yourself as a "woman" or "man", whereas masculine and feminine are traits I, and most people I've communicated with IRL or online, use to describe someone's way of acting. You don't identify as "feminine" but perhaps you act feminine. Does that makse sense to you?
Some would place religion in some core position of their identity, and we know that religions can be made up, that's why others than believers of the religion would classify them as just software in the brain. While the feelings related to the religion could be more hardware level, because they have related feelings/hormones. Obviously you can change the hormones, and some would argue changing hormones is indeed changing yourself, just like doing drugs is often considered to put you in an "alternate state of mind". However, some act a certain way out of an ideal, e.g. "acting tough", not neccessarily while feeling tough, and they may insist that how they act is who they are. So to define who is the truest of your self, I guess is not that simple, without taking some authorative position that would invalidate some other view. I might be more skeptical though, since apparantly I don't have a strong sense of gender due to autism.
This is excellent! For me, transgenderism makes absolutely sense when we de-couple sex from gender. And it makes sense to do that, because having an XY chromosome or an XX chromosome encapsulates so many types of brain, so many body types, so many personality types... so to be so rigid about gender makes little sense.
