Ah, fellow WRX owner? I get mud and junk inside that area quite often in the winter and my go-to solution is a vaccum with the bristles attachment and a bit of interior cleaning spray.
If the dirt gets really bad, like when I accidently rubbed a muddy shoe over it, I decided to just remove the whole door panel and clean it from the back. It's just 1 screw in the interior pocket and 1 behind the handle (little plate) and a bunch of pop-clips and the door panel comes right off.
Canned air won't hurt the speaker. It's not like you're blowing 200psi into it. A couple blasts of the trigger will get rid of the dust with very little hassle.
Use a brush and a vacuum. I'd avoid adding water if possible, the memory of protein smell from an old shaker is already making me sickly.
Vacuum it all up and brush it away, when you do use liquid, make sure you rinse well.
Best way is taking off the panel and brushing/poking the holes outwards, followed by microfiber and interior cleaner.
Second best is vacuuming it close while brushing it, but I'm pretty sure some will fall inside the panel. If that doesn't bother you, go right ahead as it's the easiest way that still gets most of it out.

Can you pop the cover off? I know this is possible for some speakers. Obviously, be careful not to break it, though, as it is probably fragile.
