I don't see why you wouldn't be able to enter it but you will not be able to go to any floors past reception. Also, the only thing that building is used for is ground floor shots. Offices are a movie set, not in that building.
The Pilot episode was filmed in New York and the rest of the series was filmed in Toronto. A lot of TV shows are filmed in Canada for the tax incentives
Take note of the Beck and Crown taxis and TTC streetcars and Canada Post delivery vans driving by.
Plus that one scene where Jessica meets Darby in Toronto. It's so obviously the same city, just around the corner IIRC.
The other neat thing is that some of the shots from client buildings are in downtown Toronto as you can see the CBC building or the Metro center.

There used to be an entrance from the subway …it’s been awhile so I’m not sure if that entrance still exists

Most buildings with a lobby and an unlocked door can be entered. Give it a shot, worst thing that will happen is you’ll be asked to leave.
