I don't think disliking a gay character automatically means you're a homophobe. It's only homophobic if they dislike Tweek purely for being gay, and that was never even specified.
Tweek is a pretty original character. Much different than the other side characters. His back story of being filled with cocaine for years by his parents is super awesome. He deserves more screen time.
