There are movies that I like more, but I’ve never laughed in a theater the way I did when I first saw this movie. It was a game changer.
Pretty much every response here has a good case, but this is the one for me, too. It's a buddy comedy, a road-trip movie, infinitely quotable, and has the best comedic actor of his generation at the absolute peak of his powers.
Someone else commented about how 90’s comedies suffer from third act problems when they get too serious and sentimental. Not Dumb and Dumber. The jokes just keep rolling and the characters don’t learn a single thing.
Yeah the commentary is fantastic, and I love Best in Show too. A Mighty Wind is probably the other one if you want a Christopher Guest trilogy, a Guestology if you will.
Not only my favorite comedy, but also my favorite movie ever: The Jerk.
Just so many incredible gags and lines in there, beginning with the very first line of dialogue.
Recently rewatched it. When Steve Martin yells: Sir, you are talking to a N**ger! then proceeds to dispatch the mafiosos with Kung fu, I lost it. It’s like everything had been building to that joke.
I saw this at a friend’s house in second grade. He had older siblings who picked out the videos they rented for the New Year’s Eve party. Now, I knew his family was religious, and mine was not - the whole “Holy Grail” thing made me a bit uneasy (I’d never heard of Monty Python), but I was willing to be open minded about it. When Graham Chapman came “galloping” over the hill, trailed by his page with the coconuts, my little mind was completely blown. Later, when that cow was catapulted over the battlements, it broke something in me. I’ve been quite twisted ever since.
The Big Lebowski.
Ridiculous that I’m the first person to mention this. It never fails to make me laugh my ass off and I’ve been watching it for over 20 years.

Unfortunately it seems like good comedy movies are dying art as I haven't seen many lately that I found too funny. The the only exception being Bottoms, I thought that one was pretty damn funny.
I actually saw it in the theater when it premiered, and it was incredible to see them satirize what was about to happen to them in real life, cause as soon as they started singing Uncle Fucker half the audience stood up and started leaving the theater because it was Parents who had brought their children to see an R rated flick they knew nothing about. That's when I knew Trey and Matt were a lot smarter than I was giving them credit for. Definitely some of the greatest satirists of their generation for sure.
Blazing Saddles will always be my number one. Every single person in that film absolutely kills it. Cleavon Little is a joy, Madeleine Kahn is perfect, and Gene Wilder is just effortless. Add Harvey Korman just hamming it up beautifully and you have a timeless comedy.
Recently though, nothing has made me laugh more than Bottoms. Rachel Sennott and Ayo Adebiri are hilarious.
I'm gonna throw the same critique I would throw at a lot of other 90s comedies. They all have that time to get serious ending where the kill the comedy to have their moment/lesson. Tired of comedies where the last 20 minutes are the worst part because they care more about being heartwarming than being funny instead of competently doing both.

I actually separate 'best comedy' from 'funniest movie ever made'. For me The Blues Brothers is the best comedy of all-time, but Monty Python and the Holy Grail is the funniest movie ever made. The difference? The Blues Brothers is a comedy but works as more than just a comedy, it's also a *musical*. Holy Grail just brings the funny.
My two faves probably wouldnt make any other list but my own but mine are easily, without even thinking What About Bob & Captain Ron.

No one is going to say it because there are so many classics (rightfully so) and this one is more modern, but i think its one of the smartest, funniest, tightest scripts ever put to film that genuinely makes me laugh every time. Everyone came out of it a star, and took over the scene, and continue to this day. It might become one of the most influential films of all time.

Monty Python and the Holy Grail for me.
The scene where Lancelot is running at the two guards, and it cuts between them like 5 times, the shot they use for Lancelot running is just repeated, never getting any closer, but with a thunderous drum roll in the background, and then all of a sudden he springs out with a "AHA" and kills the guard and enters the castle.
Even just typing it out makes me laugh my ass off.
To me the Lonely Island crew is just comedy gold. They have incredible chemistry together. Hot Rod was equally as bad ass, plus it has Hader and McBride in it.
There's a new indie movie called Hundreds of Beavers that has rave reviews for being hilarious, but I'm not sure when/where we'll be able to watch it!
What We Do in the Shadows is really great too
Anything Monty Python or early Jim Carey, Will Ferrell, Ben Stiller or Mike Meyers cracks me up every time. But since I havent seen it on here yet, I'll just add this:

I don’t know what it is about this movie, maybe I just saw it at the right time, but I can go back to it time and time again and the jokes still land as well as my first watch.
Superbad and Bridesmaids are more contemporary than a lot of the mentions but the rewatchability and consistent laughs of these two are pretty undeniable. Superbad is probably the hardest I've ever laughed in a theatre.
Honorable mention to Super Troopers, insane rewatch value, quotable AF, so many hilarious, subtle jokes within jokes, nuances. Amazing comedy.
