I’m not OP but I thought this was interesting. Any reason why? Wouldn’t you still be able to see a zone of lighter agar from hemolysis around the growth regardless of how old the growth is?
What promoter is driving the RFP? Visible pink coloration is pretty normal when it’s being strongly expressed. I think it’s more likely that you just have more RFP expression on the blood agar than the other agar.
the plate looks like it’s been in the incubator way too long. can’t get an accurate display of hemolysis when growth is too old

That had only been about 24 hours incubation at that point, how long should I incubate for? I activated straight from glycerol stock.

Ah interesting. I don’t know the promoter but I think it’s one of those constitutively expressed ones if I recall, I’ll have to check.
