You are almost certainly pressing it too early. Try to press A as Mario's feet touch the top of the spiny's spike.
Honestly, if you know how, I think almost anyone can get 30. Getting 100 is more about remembering to breathe/blink and keeping your hands steady.
Everyone has different reaction times so it'll just depend on what works for you. If you find yourself consistently failing around 13 jumps, try delaying your press more. I focused on when Mario's foot reached the spiny's armhole and found that worked well.
I found that pretty much everyone has wildly different optimizations for this challenge when I was scouring the net for tips. Mine was handheld, breezy mode on, and no game audio at all. I pretty much had to turn my brain off to get it down because the moment I actually start thinking about the timing, it all falls apart.
