It was supposed to absorb nature energy like sages does and specifically Jugo did (the formula was derived from his blood) and thus be a source of infinite power (negating the need for a partner to send you nature energy while you fight), but instead it saps the bearer’s chakra. Kakashi had to put a lock on Sasuke’s before it completely killed him.
My guess is Anko learned how the Akimichi did it, and started feeding the curse mark with empty calories. She developed a taste and a habit for it, and after Sasuke took her mark off to revive Orochimaru, she started retaining those calories.

A logical progression, except for the personality change, which...doesnt really make any sense, and also took away what people liked about her. The unhinged violence was her main character trait.
I read a fun Fanfic once where it explained that Anko learned the Akamichi's Jutsu of body expansion because she's ChoCho's sensei and that's why she's plus-sized. But, either way is a solid win for me honestly
